چکیده

Abstract Proteases derived from human pathogens can specifically cleave IgG into F(ab’)2 and Fc fragments. IdeS, an cleaving enzyme Streptococcus pyogenes, has shown clinical proof of concept, is approved for use before kidney transplantation. Due to the immunogenic nature these proteases, dosing limited by high prevalence pre-existing antibodies induction anti-drug after dosing. Therefore, mitigate impact immune system on our enzyme, we identify remove putative T B cell epitopes using Seismic’s IMPACT platform leveraging machine learning. To experimentally validate in silicopredictions, developed two complementary vitroassays intravenous immunoglobulin (IVIg) assess removal epitopes. First, how mutating wild type (WT) protease sequence impacts a competitive binding assay followed evaluating global protease. Additionally, vitroPBMC proliferation determine success epitope removal. Using flow cytometry, quantitively CD4 via bromodeoxyuridine (BrdU) incorporation. We demonstrate examples molecules designed through reduce both The assays show decrease competition as compared WT while achieving reduction IVIg binding. These data suggest that be used screen successful invisibilization interest.

In Silico Prediction of B-Cell and T-Cell Epitopes of Protective Antigen of Bacillus anthracis in Development of Vaccines Against Anthrax

Protective antigen (PA), a subunit of anthrax toxin from Bacillus anthracis, is known as a dominant component in subunit vaccines in protection against anthrax. In order to avoid the side effects of live attenuated and killed organisms, the use of linear neutralizing epitopes of PA is recommended in order to design recombinant vaccines. The present study is aimed at determining the dominant epi...

Removal of B-cell epitopes for decreasing immunogenicity in recombinant immunotoxin against B-cell malignancies.

Recombinant immunotoxin HA22, composed of an anti- CD22 Fv fragment fused to PE38, a truncated portion of Pseudomonas Exotoxin A (PE), has been developed for targeted treatment of various B-cell malignancies. As a foreign, internalized macromolecule, PE38 often induces lysosomal degradation and neutralizing antibodies to limit the efficacy of treating B-cell malignancies. The region of PE38 con...
